چکیده

Radiotherapy using gamma rays is a common modality of breast cancer treatment. The aim of this research is to investigate the biological response of the human breast cancer cell line MDAMB-231 and human peripheral blood mononuclear cells (PBMC) exposed in vitro to 60 Co irradiation at a single fraction of 10 Gy, 25 Gy and 50 Gy doses at 136,4 cGy.min rate. Cells were irradiated at room temperature by the Theratron 80 radiotherapy system. Biological response was evaluated through cellular viability using MTT assay and nucleus damages visualized by Propidium Iodide assay and electrophoresis agarose gel after gamma irradiation. Nucleus damages induced by Co irradiation were compared to damage caused by cell exposure to 10% methanol. The 50 Gy dose of irradiation did not stimulate nuclus damages at the same level as that affected by 10% methanol induction in the MDAMB-231. Further studies are necessary to understand these mechanisms in the MDAMB-231 human breast carcinoma cell line.

Effect of Myeloma-Derived Microparticles on in Vitro Proliferation and Viability of Alloimmune Human Peripheral Blood Mononuclear Cells

Background: Microparticles (MPs) are small vesicles released from the cell membrane. Accordingly, these contain active molecules to mediate biological processes, including cell proliferation and cell cycle progression. The fusion of myeloma cell lines with immunized B lymphocytes is a critical step of hybridoma technology. MPs modulate lymphoproliferation, thereby facilitating B cell expansion ...

Quantitation of genome damage and transcriptional profile of DNA damage response genes in human peripheral blood mononuclear cells exposed in vitro to low doses of neutron radiation

Background: Humans are exposed to ionizing radiation from different sources that include natural, occupational, medical, accidental exposures. Evaluation of the effect of low level of neutron exposure to human cells in vitro has important implications to human health. Attempts were made to measure genome damage, transcriptional profile of DNA damage response and repair genes in peripheral blood...

Enhancement of viability of radiosensitive (PBMC) and resistant (MDA-MB-231) clones in low-dose-rate cobalt-60 radiation therapy*

OBJECTIVE In the present study, the authors investigated the in vitro behavior of radio-resistant breast adenocarcinoma (MDA-MB-231) cells line and radiosensitive peripheral blood mononuclear cells (PBMC), as a function of different radiation doses, dose rates and postirradiation time kinetics, with a view to the interest of clinical radiotherapy. MATERIALS AND METHODS The cells were irradiat...

Conditioned Medium from Cultured Colorectal Cancer Cells Affects Peripheral Blood Mononuclear Cells Inflammatory Phenotype in Vitro

Background: Colorectal cancer (CRC) is the third most common cancer worldwide. Studies have indicated that immune cells and soluble factors play a key role in maintaining the balance between tumor-promoting inflammation and anti-tumor immunity. It has been shown that secreted cytokines from CRC cell lines could affect peripheral blood mononuclear cells (PBMCs), monocytes, and macrophages phenot...

Toxic Effect of Verapamil on Human Peripheral Blood Mononuclear Cells and BALB/c Peritoneal Macrophages, in vitro

Background & Aims: Verapamil as a calcium channel blocker has been broadly used in the treatment of many cardiovascular diseases such as hypertension and arrhythmia. Furthermore, the anti-tumor/ antiinflammatory effects of verapamil have been shown. In the present study, the cytotoxic effect of verapamil on human peripheral blood mononuclear cells (PBMCs) and BALB/c peritoneal macrophges has be...
